ELISA 酶結(jié)合物的制備:免疫酶技術(shù)(immunoenzymatic technique)又稱酶免疫測定法(enzyme immunoassay,EIA),是繼免疫熒光技術(shù)和放射免疫技術(shù)之后發(fā)展起來的又一種免疫標記技術(shù)。它是把抗原抗體的特異性反應(yīng)和酶的高效催化作用相結(jié)合而建立的。該技術(shù)通過化學方法將酶與抗體或抗抗體結(jié)合,形成酶標記物,這些酶標記物仍保持免疫學活性和酶的活性,然后將它與相應(yīng)的抗體或抗原起反應(yīng),形成酶標記復(fù)合物,結(jié)合在免疫復(fù)合物上的酶,在遇到相應(yīng)的底物時,則催化底物產(chǎn)生水解、氧化或還原等反應(yīng),而生成可溶性或不溶性有色物質(zhì)。然后根據(jù)顯色的深淺來反映待測樣品中抗原或抗體的含量,如生成的有色物質(zhì)為可溶性,則可用肉眼比色或酶標測定儀測定光密度值(OD)定性或定量;如生成的有色物質(zhì)為不溶性沉淀物,同時又是電子致密物質(zhì),則可用光學顯微鏡或電子顯微鏡識別和定位。因此,免疫酶技術(shù)是一項定位、定性和定量(敏感度可達每毫升ng~pg水平)的綜合技術(shù)。常用的酶是辣根過氧化物酶(horseradish peroxidase,HRP)和堿性磷酸酶(alkaline phosphatase,AKP)。
